Here is how procurement and formulation teams can verify strain-level evidence before acceptance.","Published","/files/c5/e4/gen_d1fda6602a66.png","
Most probiotic COAs that arrive with a quotation look complete: appearance, moisture, total viable count, microbiological data, heavy metals. The gap that matters commercially usually sits on the identification line, where the material is described only as Lactobacillus plantarum or Bifidobacterium lactis. A species name tells you which group of organism you bought; it does not tell you which strain you bought. That distinction decides whether a finished-product claim can be attributed, whether the label is defensible in markets that expect a strain designation, and whether you have any route of recourse when a supplier quietly changes what is inside the drum. The practical fix is not a larger QA department. It is a strain identity clause written into the specification and acceptance protocol before the first purchase order.

Strain-level differences are not academic. Within one species, strains can differ in acid and bile tolerance, adhesion to intestinal epithelium, behaviour during fermentation, and the immune responses they provoke in models. A claim built on a studied strain cannot be transferred to another strain of the same species, and a blend formulated around one strain's stability may behave differently after a substitution. This is also why total viable count is a weak control for multi-strain products. When several strains of the same species are present, the total count can remain on specification while the ratio between them has drifted. The number looks right; the formula no longer is.

Label and market access considerations reinforce the point. Some markets expect a strain designation or a culture collection number on pack, and a species-only description may create a labelling defect even when the organism is correctly declared. Supplier risk runs in parallel. Raw material is often traded under a species name, and a replacement strain can enter the supply chain without notification because, on paper, the identity field still reads the same. The buyer then experiences the change as unexplained variation in potency, colour, dispersibility or shelf-life, with no contractual hook to demand an explanation. Strain identity is therefore a procurement term, not only a laboratory question.

A credible COA separates identification levels clearly. Biochemical panels and 16S rRNA sequencing are useful screening tools, but they generally resolve to species, sometimes only to genus. They should not be presented as strain identity evidence. A report that stops at 16S has answered a different question from the one the specification asks. What the buyer needs is a method with strain-level resolution: whole genome sequencing, or a validated strain-specific PCR fingerprint, applied to the actual lot and evaluated against a defined reference.

For WGS, the conclusion is only as good as its documentation. The report should name the reference genome or type strain used for comparison, state the coverage achieved, describe the SNP calling and threshold rules, and identify the bioinformatics pipeline and database version. Without those elements, a headline match percentage cannot be reviewed or reproduced by a second laboratory. The same discipline applies to strain-specific PCR: primers, controls, specificity testing and interpretation rules should be visible. The culture collection number on the COA, whether ATCC, CGMCC, DSM or an equivalent, must also match the supplier's strain bank record and the number used in the technical dossier. A mismatch is a red flag, not a clerical error.

Method metadata deserve equal attention. The COA should state the test method, the limit of detection, and the accreditation status of the laboratory, for example CNAS or ISO 17025 scope for the specific test. Traceability matters too: sample receipt date, lot number, report number, and a clear link between the sample tested and the material shipped. For blends, a single species-level identification of the mixture is insufficient. Ask for per-strain identity confirmation for each component, and where the claim depends on composition, per-strain quantification or at least a documented enumeration strategy.

A COA that cannot be traced from the sampled lot to the shipped lot is a document, not evidence.

These expectations belong in the supply agreement. A workable structure is full identity confirmation on the first commercial lot, followed by a defined sampling plan for subsequent lots, with additional testing triggered by changes in fermenter, manufacturing site, contract manufacturer, raw material source or lyoprotectant system. The agreement should also create a notification duty: the supplier must inform you in writing before changing strain source, production site or the lyophilised matrix, and must support revalidation. Retention samples, held by both parties for an agreed period, make later disputes resolvable rather than rhetorical.

Nonconformance terms need to be equally concrete: what counts as a failed identity result, who pays for the confirmatory test, how replacement or credit is handled, and how recall costs are allocated if a deviation is found after release. Cost is usually the objection raised at this point. The relevant comparison is not the price of one identity test against zero, but the price of testing against the cost of a withdrawn claim, a relabelling exercise, a market recall, or a reformulation forced by an unannounced strain change. Spread across a year of production, identity verification is usually a small line item.

The common failure modes are predictable. Reports list only species or a trade name, with no deposit number and no strain-level comparison. WGS summaries omit the reference, coverage and SNP criteria, so the conclusion cannot be audited. The sample tested is a pilot or R&D powder rather than the commercial lot. Or the deposit number on the COA differs from the one in the technical file. A short screening checklist catches most of this: species and strain both named; deposit number present and consistent; method and resolution stated; reference and interpretation rules documented; laboratory accreditation and detection limit given; lot linkage and sample traceability clear; per-strain data for blends; change notification and retention sample clauses in place. Use it in the first supplier round, not after qualification. Ask which strain is in the drum, where it is deposited, how the lot was confirmed, and what happens if the strain source changes. Suppliers who answer with documents are easier to qualify; those who answer with a species name have told you something useful as well.
","Strain Identity Testing: Reading a Probiotic COA","Species names are not strain identity.
